There's an interview with him on Apple's site where he talks about using mathematics applications to work out new ways of folding paper. That helps get a base for some of the more complicated shapes then he has to work on the detailing.

It is hard to believe that this black widow paper is uncut. You would think he had to do some cutting to get so many legs out.

Now why didn't I think of that? Sounds like a shame to spoil perfectly good magazines though, unless you made printouts. I did find a diagram recently for an origami penis which would seem to be the ideal shape for something like that.
